Cre\u0219terea produc\u021biei de beta 42 amiloid \u00een creier poate duce la formarea pl\u0103cilor de amiloid; o sc\u0103dere a beta-amiloidului asociate cu niveluri crescute de proteine tau sau fosfo-tau, poate prezice apari\u021bia bolii AD. Boala Alzheimer este diagnosticat\u0103 \u00een prezent pe baza modific\u0103rilor cognitive \u0219i prin excluderea altor cauze ale acestor modific\u0103ri. De cele mai multe ori, diagnosticul este confirmat definitiv post-mortem, prin explorarea modific\u0103rilor microscopice din \u021besutul cerebral. Evaluarea microscopic\u0103 implic\u0103 identificarea num\u0103rului de pl\u0103ci amiloide precum \u0219i a celui de aglomerari specifice neurofibrilare, din substanta cerebrala. Modific\u0103rile caracteristice ale RMN sau PET) \u0219i \/ sau amiloid beta sc\u0103zut, insotite de niveluri ridicate de proteine \u200b\u200btau \u00een LCR, ajuta la stabilirea diagnosticului.<\/p>\n<p><strong><em>Informa\u021bii generale<\/em><\/strong><\/p>\n<p>Peptidele \u03b2-amiloide sunt fragmente proteolitice de 39- 44 aminoacizi, derivate din proteina precursoare amiloid\u0103 (APP), o glicoprotein\u0103 transmembranar\u0103 care este codificat\u0103 de cromozomul 21 \u0219i exprimat\u0103 \u00een majoritatea celulelor de mamifere<sup>1<\/sup>. Fragmentele de amiloid formeaz\u0103 nucleul pl\u0103cilor senile din boala Alzheimer \u0219i sunt implicate ca factor determinant major, \u00een aceast\u0103 patologie<sup>2,3<\/sup>. \u03b2-amiloid-ul<sup>1-42\u00a0<\/sup>este cel mai toxic dintre aceste specii\u00a0<em>in vitro<\/em>\u00a0\u0438\u00a0<em>in vivo<\/em>\u00a0\u0219i este componenta principal\u0103 a nucleului pl\u0103cii senile. Muta\u021biile genetice care au ca rezultat cre\u0219terea produc\u021biei de \u03b2-amiloid<sub>1-42<\/sub>\u00a0\u0219i, \u00een consecin\u021b\u0103, o apari\u021bie si evolu\u021bie accelerat\u0103 a bolii Alzheimer, ofer\u0103 dovezi c\u0103 \u03b2-amiloid<sub>1-42<\/sub>\u00a0joac\u0103 un rol central \u00een patogenia acestei tulbur\u0103ri<sup>4-6<\/sup>.
