Insomnia este o tulburare a ini\u021bierii sau men\u021binerii somnului, iar prevalen\u021ba sa la persoanele \u00een v\u00e2rst\u0103 este estimat\u0103 la 23-34%<sup>1<\/sup>. NZP func\u021bioneaz\u0103 ca un hipnotic, pentru a prelungi timpul de somn \u0219i pentru reducerea timpul necesar adormirii, dar pot ap\u0103rea mai multe efecte secundare, cum ar fi leziuni hepatice \u0219i teratogenitate. Insuficien\u021ba hepatic\u0103 colestatic\u0103 a fost raportat\u0103 ca urmare a utiliz\u0103rii NZP. Se consider\u0103 c\u0103 aceste reac\u021bii adverse sunt induse prin metabolismul NZP<sup>2,3<\/sup>.<\/p>\n<p>La om, NZP este metabolizat \u00een principal \u00een 7-amino nitrazepam (ANZP) de nitro-reductaz\u0103 (s) \u0219i ulterior \u00een 7-acetilamino nitrazepam (AANZP) de N-acetiltransferaz\u0103 (s)<sup>4<\/sup>. \u00cen termen de 24 de ore de la administrarea oral\u0103 de 10 mg NZP la om, excre\u021bia urinar\u0103 de ANZP \u0219i AANZP a reprezentat 4.1% \u0219i, respectiv, 12.5% din doza ini\u021bial\u0103. Al\u021bi posibili metaboli\u021bi NZP au fost detecta\u021bi \u00eentr-o m\u0103sur\u0103 mult mai mic\u0103 \u00een urina uman\u0103, comparativ cu ANZP \u0219i AANZP<sup>1<\/sup>.<\/p>\n<p>Toxicitatea NZP a fost evaluat\u0103 prin studii in vitro.
