Intr-o celula in care se acumuleaza radicalii liberi, acizii grasi, care in mod normal ar fi supusi \u03b2-oxidarii in mitocondrii, vor urma calea peroxidarii lipidice in urma careia rezulta ca produs final malondialdehida (MDA).<\/span><\/p>\n<p><span style=\"font-size: medium;\">Malondialdehida poate fi intalnita in multe probe biologice (ser, plasma, urina, tesuturi si chiar produse alimentare) si a devenit unul dintre cele mai folositi indicatori pentru a estima efectele stresului oxidativ asupra lipidelor<sup>2;5<\/sup>.<\/span><\/p>\n<p><span style=\"font-size: medium;\">Este o dialdehida (alcatuita din 3 atomi de carbon) intens reactiva, formata in toate celulele de mamifere in urma procesului de peroxidare a acizilor grasi polinesaturati si din metabolismul acidului arahidonic (biosinteza prostaglandinelor si tromboxanului). Se leaga covalent la gruparile amino ale proteinelor, lipoproteinelor, fosfolipidelor sau ADN-ului. MDA reactioneaza cu ADN-ul pentru a forma malondialdehid-deoxiguanozine si deoxiadenozine, insa compusul principal este o pirimidopurinona, numita M1G, care poate contribui semnificativ la aparitia cancerului determinat de stilul de viata si factorii dietetici.<\/span><\/p>\n<p><span style=\"font-size: medium;\">Spre deosebire de radicalii liberi, aldehidele sunt relativ stabile si, prin urmare, pot sa difuzeze prin membrane si pot astfel actiona la distanta de locul original al radicalilor liberi<sup>1;4;6<\/sup>.<\/span><\/p>\n<p><span style=\"font-size: medium;\">Proteinele modificate pot prezenta alterari structurale, ale antigenitatii si comportamentului fizico-chimic (disfunctie sau inactivare). Desi MDA poate fi inactivata de aldehid-dehidrogenaze, productia sa este accelerata de stresul oxidativ si, atunci cand concentratiile ating niveluri critice, dialdehida poate scapa procesului de detoxifiere. MDA este un compus toxic implicat in procesul de mutageneza la persoanele varstnice, carcinogeneza, nefropatie diabetica, leziuni determinate de radiatii si o serie de alte procese patologice.
